Base Degradation Monitoring of Dolasetron Mesylate by UV Spectrophotometric Method

A simple and fast stability indicating spectrophotometric method has been developed and validated for estimation of Dolasetron mesylate that can be used for routine analysis. Dolasetron mesylate is a potent antinauseant and antiemetic agent. It is a serotonin 5-HT3 receptor antagonist. Spectrophotometric detection of Dolasetron mesylate shows maximum absorbance at 285 nm. Alkali degradation monitoring of Dolasetron mesylate was performed by first derivative spectrophotometric method at 229 nm. The method was found to be linear in concentration range 10-100μg/ml. Mean assay of Dolasetron mesylate was found to be 101.48%. The method validated for parameters such as linearity, range, precision, robustness, sensitivity and accuracy according to ICH Q2R1 (International Conference on Harmonization). When stress degradation studies were carried under ICH recommended stress condition of acid/ base/ neutral hydrolysis, oxidation, dry heat and photolysis according to ICH Q1AR2, it was observed that presence of Dolasetron degradation product could be detected by first derivative UV method.
